First there is the tumor and then there's the horrible question of whether the cancerous cells will spread. Scientists increasingly believe that the structural properties of the tumor itself, such as how tightly the tumor cells are packed together, play a decisive role in the progression of the disease. In a new study, researchers show that the protein Pannexin1, known to have tumor-suppressive properties, plays an important role in keeping the cells within a tissue closely packed together, an effect that may be lost with cancer.
"In healthy tissues, the recently discovered protein Pannexin1 may be playing an important role in upholding the mechanical integrity of the tissue," said first author and Brown University M.D./Ph.D. student Brian Bao. "When we develop cancer, we lose Pannexin1 and we lose this integrity."
The results appeared in advance online in the Journal of Biological Chemistry on Jan. 20.
To conduct their research, the group at Brown University and the University of British Columbia employed a "3-D Petri dish" technology that allows investigators to watch closely how cells interact with each other, without scientists having to worry about additional interactions with surrounding scaffolding or the culture plate itself. How readily the cells form large multicellular structures therefore reflects their interactions with each other, not their in vitro surroundings.
Bao's advisor, Jeffrey Morgan, associate professor of medical science, developed the 3-D Petri dish technology. Morgan is the paper's senior author.
Cancer cells converge
Starting with rat "C6" glioma (brain tumor) cells that do not express Pannexin1, the researchers left some unaltered and engineered others to express Pannexin1. After putting the different cells into the 3-D Petri dishes and watching them interact for 24 hours, they saw that the Pannexin1 cells were able to form large multicellular tissues much faster and more tightly than the unaltered cancer cells.
To confirm that Pannexin1 was indeed causing these changes, Bao and his colleagues treated their samples with the drugs Probenecid and Carbenoxolone, which are well known inhibitors of Pannexin1. They saw that sure enough, the drugs negated Pannexin1's accelerating effect.
Then the team was ready to achieve the the study's main aim, Bao said, namely to determine how Pannexin1 was able to drive these cells to clump together faster and tighter. They found that Pannexin1 sets off a chain reaction involving the energy-carrying molecule ATP and specific receptors for it.
When all experiments were done, Bao, Morgan, and their collaborators had found that as soon as the cells touched each other, Pannexin1 channels were stimulated to open and release ATP. The ATP then bound to cell surface receptors, kicking off intracellular calcium waves that ultimately remodeled the network of a structural protein called actin. This remodeling increases the forces between the cells, driving them to bind together more tightly.
Figuring out that sequence, and Pannexin1's role in it, is perhaps the study's biggest contribution to cancer research, Bao said.
"Using their single-cell systems, others have been able to carefully study individual pieces of this cascade," he said. "We came from a different perspective. Because the strength of our assay is that we can look at gross multicellular behavior in 3-D, we could ask, 'Does this actually manifest into something tangible on the multicellular level?'"
Having gained this understanding of Pannexin1's role in the mechanics of tumors, Bao is now engaged in research to answer the obvious next questions: Does Pannexin1 affect the tumor's ability to spread and invade? When cancerous cells regain Pannexin1 expression, are they less likely to spread and leave the tumor?

CAIRO -- A professor from American University in Cairo says discovery of prostate cancer in a 2,200-year-old mummy indicates the disease was caused by genetics, not environment.
The genetics-environment question is key to understanding cancer.
AUC professor Salima Ikram, a member of the team that studied the mummy in Portugal for two years, said Sunday the mummy was of a man who died in his forties.
She said this was the second oldest known case of prostate cancer.
"Living conditions in ancient times were very different; there were no pollutants or modified foods, which leads us to believe that the disease is not necessarily only linked to industrial factors," she said.
A statement from AUC says the oldest known case came from a 2,700 year-old skeleton of a king in Russia.

BEIJING (AP) ? A young man posts his photo with a leaflet demanding freedom for Tibet and telling Chinese police, come and get me. Protesters rise up to defend him, and demonstrations break out in two other Tibetan areas of western China to support the same cause.
Each time, police respond with bullets.
The three clashes, all in the past week, killed several Tibetans and injured dozens. They mark an escalation of a protest movement that for months expressed itself mainly through scattered individual self-immolations.
It's the result of growing desperation among Tibetans and a harsh crackdown by security forces that scholars and pro-Tibet activists contend only breeds more rage and despair.
That leaves authorities with the stark choice of either cracking down even harder or meeting Tibetan demands for greater freedom and a return of their Buddhist spiritual leader, the Dalai Lama ? something Beijing has shown zero willingness to do.
"By not responding constructively when it was faced with peaceful one-person protests, the (Communist) party has created the conditions for violent, large-scale protests," said Robbie Barnett, head of modern Tibetan studies at New York's Columbia University.
This is the region's most violent period since 2008, when deadly rioting in Tibet's capital Lhasa spread to Tibetan areas in adjoining provinces. China responded by flooding the area with troops and closing Tibetan regions entirely to foreigners for about a year. Special permission is still required for non-Chinese visitors to Tibet, and the Himalayan region remains closed off entirely for the weeks surrounding the March 14 anniversary of the riots that left 22 people dead.
Video smuggled out by activists shows paramilitary troops equipped with assault rifles and armored cars making pre-dawn arrests. Huge convoys of heavily armored troops are seen driving along mountain roads and monks accused of sedition being frog-marched to waiting trucks.
For the past year, self-immolations have become a striking form of protest in the region. At least 16 monks, nuns and former clergy set themselves on fire after chanting for Tibetan freedom and the return of the Dalai Lama, who fled to India amid an abortive uprising against Chinese rule in 1959.
China, fiercely critical of the Dalai Lama, says Tibet has been under its rule for centuries, but many Tibetans say the region was functionally independent for most of that time.
In a change from the individual protests, several thousand Tibetans marched to government offices Monday in Ganzi prefecture in Sichuan province. Police opened fire into the crowd, killing up to three people, witnesses and activist groups said.
On Tuesday, security forces opened fire on a crowd of protesters in another area of Ganzi, killing two Tibetans and wounding several more, according to the group Free Tibet.
On Thursday in southwestern Sichuan province's Aba prefecture, a youth named Tarpa posted a leaflet saying that self-immolations wouldn't stop until Tibet is free, the London-based International Campaign for Tibet said. He wrote his name on the leaflet and included a photo of himself, saying that Chinese authorities could come and arrest him if they wished, group spokeswoman Kate Saunders said in an email.
Security forces did so about two hours later. Area residents blocked their way, shouting slogans and warning of bigger protests if Tarpa wasn't released, Saunders said. Police then fired into the crowd, killing a a 20-year-old friend of Tarpa's, a student named Urgen, and wounding several others.
The incident, as with most reported clashes in Tibetan areas, could not be independently verified and exact numbers of casualties were unclear because of the heavy security presence and lack of access. The topic is so sensitive that even government-backed scholars claim ignorance of it and refuse to comment.
The government, however, acknowledged Tuesday's unrest, saying that a "mob" charged a police station and injured 14 officers, forcing police to open fire on them. The official Xinhua News Agency said police killed one rioter and injured another.
"The Chinese government will, as always, fight all crimes and be resolute in maintaining social order," Foreign Ministry spokesman Hong Lei said in comments on the incident.
The harsh response points to a deep anxiety about the self-immolations, said Youdon Aukatsang, a New Delhi-based member of the Tibetan parliament-in-exile.
"They're worried that there is an underground movement in Tibet that is coming to the surface," she said.
Tibetan desperation has been fed both by the harsh crackdown ? security agents reportedly outnumber monks in some monasteries ? along with a deep fear that the Dalai Lama, probably the most potent symbol of Tibet's separate identity, will never return.
The 76-year-old Nobel Peace Prize laureate handed his political powers to an elected assembly last year. That was intended to ensure the Tibetan cause would live on after him, but was met with considerable anxiety among many Tibetans who saw it as a sign he was giving up his role as leader of their struggle.
Dibyesh Anand, a Tibet expert at London's University of Westminster, said resistance to Chinese rule is likely to grow more fierce.
"Protests will get more radicalized since the Tibetans in the region see no concession, no offer of compromise, no flexibility coming from the government," he said.

UNITED NATIONS ? A shipment containing 16 kilograms of cocaine was seized last week at the U.N.'s mail intake center, a New York Police Department spokesman said Thursday.
Paul Browne, NYPD's chief spokesman, said the drug was in a white bag evidently masquerading as a diplomatic pouch that raised suspicions when it was being scanned because it was stamped with what looked like a poorly concocted version of the U.N. logo.
Browne said there was no name or address on the shipment sent from Mexico City through Cincinnati.
U.N. security officials called the NYPD and Drug Enforcement Administration, which confirmed the substance inside the shipment intercepted Jan. 16 was cocaine, the police spokesman said.
U.N. undersecretary-general for safety and security Gregory B. Starr told reporters Thursday evening that "there is nothing to indicate that this had anything to do with anybody at the United Nations."
Starr said the drug was actually stashed in two bags that were stamped with the sky-blue U.N. logo of a world map in an apparent effort to masquerade as diplomatic pouches, which are not supposed to be inspected. Inside the bag, the drug was hidden in hollowed-out notebooks, he added.
The U.N. official showed journalists a photograph of the bags that were seized, and compared them with a real diplomatic pouch used by the U.N., which is somewhat larger and made of a different material.
"This did not come from a United Nations facility," Starr said of the shipment. "It was not, in my opinion, not intended to go to a United Nations facility."

As Tea Party state education chief John Huppenthal retreats into his office after an embarrassing national media tour on Arizona's extremist Ethnic Studies crackdown, and Tucson Unified School District administrators continue their slide into a public relations disaster over banishing Mexican American Studies curricula and books, a remarkably diverse array of librarians, educators, writers, civil rights activists and the Congressional Hispanic Caucus is mounting a series of national actions to call attention to educational and civil rights violations and to support local Tucson efforts.
On January 24th, the American Library Association issued a condemnation of Arizona's "suppression of open inquiry and free expression caused by closure of ethnic and cultural studies programs on the basis of partisan or doctrinal disapproval," and the Tucson school district's "restriction of access to educational materials associated with ethnic and cultural studies programs." The national library association, with active chapters across the country, also called on the state to support a new bill to repeal the Ethnic Studies ban.
As a follow up to their extraordinary request to the Civil Rights Division of the Department of Justice and the Department of Education this week for a federal investigation of civil rights violations by the state of Arizona, the Congressional Hispanic Caucus is urging constituents to change their profile picture on Facebook and Twitter to a special logo -- "You Can't Ban Books, You Can't Ban History" -- on Thursday, January 26, 2012.
On February 1st, teachers and schools around the country have been encouraged by Rethinking Schools, whose nationally acclaimed textbook Rethinking Columbus: The Next 500 Years was confiscated and banished from Tucson schools, to follow the suggestion of former Tucson Mexican American Studies literature teacher Curtis Acosta for a "national day of solidarity where teachers would teach our curriculum all over the nation."
Along with special forums planned across the country, from California to New York, a network of educators in Georgia is sponsoring a "Teach-in" in Atlanta on Saturday, Feb. 4th.
The event is framed as a "Teach-in," where we can inform the community about what's happening, work together to fight censorship and racism in schools, and make plans for future social justice activism. Groups will include:
(1) curricular action, in which participants create lesson plans and activities for PK-12 students on issues of censorship, critical pedagogy, and/or Mexican American history;
(2) censored books dialogue, in which participants learn about the books that were banned and the theories contained within them; and
(3) legislative overview, in which participants discuss legal implications of the ban in Arizona and around the country.
Finally, the group will come back together to plan action steps that can be taken in higher education, PK-12 schools, and communities in Georgia and around the country.
Several national petitions are also being circulating, including a change.org petition by former Mexican American Studies teacher Norma Gonzales, who has called on the Tucson school district to "immediately remove these books from their 'district storage facility' and make them available in each school's library. Knowledge cannot be boxed off and carried away from students who want to learn!"
In a stunning revelation last week, a review of the TUSD library catalog found that there are less than 2 or 3 copies of some of the banished texts in libraries serving more than 60,000 students.
Presente.org, the national Latino and human rights organization, is also circulating a petition to "tell Superintendent Pedicone and the school board to reverse the ban and reinstate the Mexican American Studies program."
In one of the most creative actions to take on Arizona's removal of books and texts, Texas author and literary organizer extraordinaire Tony Diaz is assembling a caravan of renowned authors and librotraficantes to deliver banished books to Arizona students in March.

BANGKOK ? Oil rose above $100 a barrel Thursday in Asia after the U.S. Federal Reserve said it would keep interest rates at record lows at least until 2014 to help jump-start the world's biggest economy.
Benchmark crude for March delivery was up 71 cents at $100.11 a barrel at midday Bangkok time in electronic trading on the New York Mercantile Exchange. The contract rose by 45 cents to finish at $99.40 per barrel in New York on Wednesday. At one point it was as high as $100.40.
Brent crude for March delivery was up 88 cents at $110.69 a barrel on the ICE Futures Exchange in London.
The U.S. central bank, which has kept its benchmark interest rate near zero for three years, said Wednesday that it doesn't plan to raise the rate before late 2014.
That caused the dollar to turn lower against major currencies, which makes dollar-priced oil less expensive for holders of other currencies.
"That would mean the U.S. dollar would continue to be cheap versus other currencies, and there is typically an inverse correlation between the value of the dollar and commodity pricing," said Victor Shum, an energy analyst at consultancy Purvin & Gertz in Singapore.
"So oil prices are supported by the Federal Reserve statement," he said.
But other analysts saw room for oil prices to fall.
Leaving rates low would encourage businesses and consumers to borrow money cheaply, boosting the economy and leading to higher oil demand. But the Fed also "telegraphed its concern regarding U.S. economic growth ... which is intuitively bearish for oil," said energy trader and consultant The Schork Group.
In other Nymex trading, heating oil rose 2.8 cents to $3.04 per gallon and gasoline futures gained 1.7 cents at $2.86 per gallon. Natural gas advanced 2.4 cents to $2.75 per 1,000 cubic feet.

PARK CITY, UTAH (Reuters) - Filming "The Hobbit" has seemed like a family get together of sorts for director Peter Jackson and at least one of his "Lord of the Rings" stars, Elijah Wood.
"I'm enjoying it much more than I ever thought I would and it literally does feel like a family reunion, which is a nice way to go to work each day," Jackson told Reuters at the Sundance film festival on the red carpet to promote his documentary film, "West of Memphis."
Wood, also at Sundance but for a separate film in which he stars, "Celeste and Jesse Forever," told Reuters that the "Hobbit" shoot was "pretty extraordinary."
"It was very surreal," Wood said. "It kind of felt like no time had passed and suddenly we were back working again. It kind of felt like stepping back into time, family reunion. It was so great."
"The Hobbit," based on J.R.R. Tolkien's fantasy novel, is a two-part prequel to Jackson's "Lord of the Rings" trilogy, which saw Wood in the starring role of Frodo Baggins. The movies became among the biggest blockbusters ever, and the final film, "The Lord of the Rings: The Return of the King," won 11 Oscars including best film and best director for Jackson.
"Hobbit" audiences will see many of the same faces reprising their characters from the "Rings" films, including Wood, Ian McKellen as Gandalf, Andy Serkis as Gollum, Orlando Bloom as Legolas and Cate Blanchett as Galadriel.
Wood said his part is "really small" and joked that when he finished shooting his scenes, he wished didn't have to leave. "I was telling Andy Serkis, who's (also) directing second unit, that I wanted to stow away on (his) unit and keep working," said Wood.
Newcomers to the 'Hobbit' cast include Martin Freeman as Bilbo Baggins and Richard Armitage as Thorin Oakenshield. Luke Evans, Evangeline Lilly and Lee Pace also are among the cast. The story follows Bilbo on an epic quest with 13 dwarves and the
warrior Thorin to reclaim the Dwarf Kingdom of Erebor.
Part one, "An Unexpected Journey," is scheduled for a December 2012 release while part two, "There and Back Again," will land in movie theaters one year later.

After 10 hours of talks on Monday, the finance ministers of the countries that use the euro announced that Greece would pay less than 4 percent interest on the new bonds creditors will get in a swap meant to cut Greece's debt by about euro100 billion ($130 billion).
The deal is crucial to Greece's and the eurozone's stability since it's clear there's no way Athens can ever pay back all that it owes. Banks that hold Greek debt have already been asked to take a 50 percent loss on those investments ? and some think even that writedown isn't big enough.
The negotiations involve a delicate balancing act between getting a deal large enough to ensure that Greece can someday dig out from under its pile of debts but not so harmful to banks that it scares investors off from investing in any eurozone debt.
European leaders have promised Greece is a special case and bondholders won't ever be asked to take losses again, but there are signs that investors are staying clear of the bonds of other vulnerable countries, like Portugal.
Time is running out for politicians and the banks to get it right ? Greece has several billions of euros of debt coming due in March ? and stocks dropped Tuesday amid worries they might not.
In France, the CAC-40 fell 0.8 percent to 3,312, while Germany's DAX dropped 1 percent at 6,370. The FTSE index of leading British shares was down 0.7 percent to 5,740.
Wall Street was also set to open lower. Dow futures fell 0.3 percent at 12,609 and S&P futures dropped 0.4 percent to 1,305.
The euro fell 0.2 percent to $1.3000.
Politicians are also aware that European banks are under tremendous pressure because of the amount of government debt they hold and have seen their stock prices crash and their sources of funding dry up during the crisis. Late Monday, Standard & Poor's downgraded the credit ratings of two major French banks, Credit Agricole and Societe Generale. They confirmed the rating of a third major bank, BNP Paribas, but the stock prices of all three plummeted Tuesday, underscoring how fragile all financial institutions are.
Compounding these concerns is the poor state of Europe's economy and worries that the eurozone is slipping back into recession. Even relatively positive results from two economic surveys released Tuesday were not enough to ease those worries.
January's manufacturing purchasing managers' composite index rose to 48.7 from 46.9, according to Markit, a financial data company. The services PMI rose to 50.5 from 48.8. Both surveys, which are considered indicators for growth, beat the expectations of analysts, but experts warned they are far from good news.
"While the January purchasing managers' surveys lift hopes that eurozone activity is stabilizing, they also suggest that the eurozone is far from out of the economic woods," said Howard Archer, an analyst with IHS Global Insight. "Worrying elements remain in the purchasing managers' surveys and we suspect that it is still more likely than not that the eurozone will suffer further contraction in the first quarter of 2012 which will put it back into recession."
Concerns about the state of economy even tempered oil prices, which had been skyrocketing after European leaders announced they would stop buying Iranian oil in an effort to pressure Tehran into resuming talks on its nuclear program.
Benchmark oil fell back 30 cents to $99.28 in electronic trading on the New York Mercantile Exchange.
Earlier in Asia, Japan's Nikkei 225 stock rose 0.2 percent to 8,785.33 despite the central bank cutting growth forecasts for the fiscal year ending March 2012 and the following year because of a slowdown in overseas demand and the strong yen.
Australia's S&P/ASX 200 closed little changed at 4,224.20. Indonesia's benchmark was up 0.1 percent at 3,994.91 and India's Sensex was 1.5 percent higher at 16,997.35 after the Reserve Bank of India lowered cash reserve requirements for commercial lenders.

ScienceDaily (Jan. 15, 2012) ? Scientists working at the Medical Research Council have identified changes in the patterns of sugar molecules that line pre-cancerous cells in the esophagus, a condition called Barrett's dysplasia, making it much easier to detect and remove these cells before they develop into esophageal cancer. These findings, reported in the journal Nature Medicine, have important implications for patients and may help to monitor their condition and prevent the development of cancer.
Esophageal cancer is the fifth biggest cause of cancer death in the United Kingdom and the eighth leading cause of cancer deaths for men in the United States. Moreover, the number of people diagnosed with this disease is increasing rapidly. Individuals with a pre-cancerous condition known as Barrett's esophagus are at an increased risk of developing esophageal cancer, and need to be closely monitored to make sure that the disease is not progressing.
Dysplasia offers a stage at which cancer can be prevented by removing these cells. However correctly identifying these areas has proved to be problematic, as they can easily be missed during endoscopy and biopsy, which only take samples from a small part of the esophagus. This can result in false reassurance for patients in whom their dysplasia has been missed, and conversely those without dysplasia having to undergo further unnecessary treatments.
The team, based at the MRC Cancer Cell Unit in Cambridge, was led by Dr. Rebecca Fitzgerald and included New York University's Lara Mahal, an associate professor of chemistry, and William Eng, a laboratory technician.
The researchers discovered a new mechanism for identifying Barrett's dysplasia cells by spraying on a fluorescent probe that sticks to sugars and lights up any abnormal areas during endoscopy. By analyzing the sugars present in human tissue samples taken from different stages on the pathway to cancer -- using microarray technology developed by NYU's Mahal -- they found that there were different sugar molecules present on the surface of the pre-cancerous cells. This technology uses sugar binding proteins, known as lectins, to identify changes in sugars and pinpointed carbohydrate binding wheat germ proteins as a potential diagnostic. When the wheat germ proteins, attached to a fluorescent tag that glows under a specific type of light, were sprayed onto tissue samples, it showed decreased binding in areas of dysplasia, and these cells were clearly marked compared with the glowing green background.
"The rise in cases of esophageal cancer both in the UK and throughout the Western world means that it is increasingly important to find ways of detecting it as early as possible," Fitzgerald said. "Our work has many potential benefits for those with Barrett's esophagus who have an increased risk of developing esophageal cancer."
"We have demonstrated that binding of a wheat germ protein, which is cheap and non-toxic, can identify differences in surface sugars on pre-cancerous cells," she added. "And when coupled with fluorescence imaging using an endoscopic camera, this technique offers a promising new way of finding and then treating patients with the highest risk of developing esophageal cancer, at the earliest stage."

Local authorities were fielding inquiries from dozens of nations worried about the 4,234 people who were aboard the ship when it ran aground and tipped over off the coast of Italy, including Italians, Germans, French, Britons and Americans, and about 1,000 crew members from across the globe.
As international travel has grown easier, aid agencies and lawmakers alike have frequently warned of the potential for confusion in the wake of international emergencies, as sometimes competing nations or international organizations arrive at a disaster site.
In Italy, a host of countries sent diplomatic staff to the scene as three bodies were recovered from the sea off the tiny island of Giglio, close to the coast of Tuscany.
British ambassador Christopher Prentice said he had seen his counterparts from Germany and Spain at local hospitals, where diplomats were checking identities and tallying numbers of those injured.
Officials from the U.K. and Australia set up a joint base at Porto Santo Stefano middle school, which had been transformed into a temporary holding center for rescued passengers.
Though the school was a scene of chaos as passengers tried to find buses to take them back to Rome or the coastal town of Savona and embassy officials cross-referenced ship logs and passenger lists, Prentice said nations were cooperating well.
"This is obviously a very serious and major incident. My impression at this stage is that the Italian authorities have responded excellently and our cooperation with them has been very good," Prentice said, as he offered advice to Britons at the school.
Other embassies sent lower-level officials to work with the ship operator Costa and local authorities, offering help to foreign passengers who didn't speak Italian and were struggling to understand the response to the accident or how to get home.
Consular officials wore bright green or orange emergency vests to identify themselves to their co-nationals, offering help in how to obtain emergency passports, since many non-Europeans had to turn them in to cruise officials upon boarding.
Prentice said that in a still unfolding crisis scenario, good coordination was key. "It is about cooperation, and things are being done here very calmly and sensibly, I've been impressed by the effort of the Italian authorities," he said.
As nations were still attempting to confirm the identities of passengers who had been rescued from the ship, rescuers focused on several dozen people still unaccounted for.
Monty Mathisen, of the New York-based publication Cruise Industry News, said Costa would be well prepared to handle the demands of countries searching for news of the passengers and crew.
"They are well set up to deal with those kind of issues," said Mathisen. "The cruise industry is one of most regulated industries."
Marcus Oxley, then disaster management director of the relief charity Tearfund told a committee of British lawmakers in 2006 of the nightmare confronting local authorities as organizations descend on an area requesting information, or offering help.
"In the white hot heat of an emergency, these things are extraordinarily difficult to do," he said.

BEIJING ? Angry customers and gangs of scalpers threw eggs at Apple Inc.'s flagship Beijing store Friday after its opening for the China launch of the iPhone 4S was canceled due to concerns over the size of the crowd.
Apple reacted to the scuffle by postponing iPhone 4S sales in its mainland China stores to protect the safety of customers and employees. It said the phone still will be sold online and through its local carrier.
Customers including migrant workers hired by scalpers in teams of 20 to 30 to buy iPhones for resale at a markup to Chinese gadget fans waited overnight in freezing weather at the Apple store in Beijing's east side Sanlitun district.
The crowd erupted after the store failed to open on schedule at 7 a.m. Some threw eggs and shouted at employees through the windows.
A person with a megaphone announced the sale was canceled. Police ordered the crowd to leave and sealed off the area with yellow tape. Employees posted a sign saying the iPhone 4S was out of stock.
"We were unable to open our store at Sanlitun due to the large crowd, and to ensure the safety of our customers and employees, iPhone will not be available in our retail stores in Beijing and Shanghai for the time being," said Apple spokeswoman Carolyn Wu.
Other Apple stores in China opened Friday and Wu said iPhone 4S quickly sold out. She said the phone still will be sold in China through Apple's online store, its local carrier China Unicom Ltd. and retailers that are authorized resellers.
Wu declined to comment on what Apple might know about scalpers buying iPhones for resale.
China is Apple's fastest-growing market and "an area of enormous opportunity," CEO Tim Cook said in October. He said quarterly sales were up nearly four times over a year earlier and accounted for one-sixth of Apple's global sales.
Apple's iPhones are hugely popular in China and stores are mobbed for the release of new products.
iPhones are manufactured in China by an Apple contractor but new models are released in other countries first. That has helped to fuel a thriving "gray market" in China for phones smuggled in from Hong Kong and other markets.
Last May, the Sanlitun store was closed for several hours after a scuffle between an employee and a customer during the release of the iPhone 4, the previous model in the series.
Customers began gathering Thursday afternoon outside the Sanlitun store. People in the crowd said the number grew to as many as 2,000 overnight but many left before dawn after word spread that the store opening would be cancelled. There were about 350 people left when the protest erupted after 7 a.m.
"On the one hand there is poor organization and on the other there were just too many people," said a man outside the Sanlitun store Friday, who would give only his surname, Miao. "I don't think they prepared well enough."
Another man who refused to give his name said he was a migrant laborer who was paid 100 yuan ($15) to wait in line overnight.
Others in the crowd said scalpers had organized groups of 20 to 30 migrant workers to buy phones or hold places in line. Organizers held colored balloons aloft to identify themselves to their workers.
Others said they were waiting to buy the phone for themselves.
"I just like the 4S," said Zhu Xiaodong, a Beijing resident. He said he was upgrading from the previous iPhone 4 model.

Last week started badly for Inland Assemblyman Tim Donnelly and only got worse.
The Twitter-verse erupted Tuesday after security officers found a loaded handgun in Donnelly?s carry-on luggage at Ontario International Airport. Donnelly, R-Twin Peaks, said he forgot he had put the gun in his briefcase and believed he was carrying his laptop computer.
Within hours, someone had created a ?TimDonnellysGun? Twitter account. ?Guns don?t kill, they?re like laptops,? wrote the unidentified author, who had 115 followers by Friday.
Big Bear Lake Mayor Bill Jahn, a Republican who is challenging Donnelly in the redrawn 33rd Assembly District, piled on.
?I?m a gun owner, too, but I know where my gun is at all times,? said Jahn, who reported last week that he had given almost $100,000 to his campaign.
The week ended with the failure of a Donnelly led campaign to qualify a ballot measure to overturn last year?s law making scholarship aid available for college students who are in the country illegally.
?Today only marks the end of one battle in a war to reclaim our voice in our legislature,? Donnelly wrote
UCR ?WASTE? DISPUTED
A study at UC Riverside that?s trying to gauge the enjoyment people get from social media is making Sen. Tom Coburn an unhappy man.
Coburn, R-Okla., is a frequent critic of what he calls wasteful government spending. And just like Santa, he makes his naughty list known in December, with the release of his annual Wastebook report that identifies the 100 most troubling uses of federal money.
?Video games, robot dragons, Christmas trees, and magic museums. This is not a Christmas wish list, these are just some of the ways the federal government spent your tax dollars,? Coburn said in a news release.
Two Inland projects made the list. UCR researchers are using $149,900 from a National Science Foundation grant to develop a video game for phones and other mobile devices that teaches children the evolutionary development of guppies. The game partners university staffers with private video game companies and includes a social media campaign, Coburn said.
Another UCR project also trades in tweets.
Two researchers were awarded $413,756 over two years to examine whether use of social media sites like Twitter and Facebook make people happy, Coburn said.
The university defended the projects, pointing out that both are led by prominent researchers in their field.
?Since hundreds of millions of people use social media it is important to know whether this helps or hurts psychological functioning,? UCR spokesman Sean Nealon said, in an email.
?If people read the proposals,? Nealon said, ?they will see the full picture and the reasons why these subjects are among many that are important to study.?
EAR FOR AN EAR
Inland Rep. Mary Bono Mack and Republican presidential frontrunner Mitt Romney have forged a friendship over the past few years.
Romney was there for the Palm Springs Republican?s last congressional bid, headlining a Riverside County fundraiser. And Bono Mack was among Romney?s earliest supporters when he announced his 2012 candidacy.
She had his back again in recent weeks, when the Romney camp was signaling it might opt to focus on this week?s New Hampshire primary instead of trying to win the Iowa caucus.
But Bono Mack advised him not to give up on the Hawkeye State, according to her spokesman, Ken Johnson.
?She personally told him and his team that they needed to spend more time in Iowa,? Johnson said.
Of course, Romney went on to eke out a narrow victory in Iowa. And if he wins the GOP nomination and beats President Barack Obama in November, the bond between Romney and Bono Mack could benefit the Inland region, Johnson said.
?Congresswoman Bono Mack certainly has his ear,? he said.
A FRIENDLY AUDIENCE?
The Service Employees International Union Local 721 plans a town hall meeting with the three candidates seeking the District 1 seat on the Riverside County Board of Supervisors.
Incumbent Bob Buster faces challenges from Assemblyman Kevin Jeffries, R-Lake Elsinore, and retired CHP officer Mike Soubirous.
The forum is billed as an endorsement town hall, but SEIU Local 721 spokeswoman Tracy Silveria said the meeting is closed to the media. It?s for SEIU members only, she said by email.
All three candidates have been invited. But will Buster have a chance at the endorsement if he attends? History might say no.
Buster has been at odds with the union as he has pushed for employees to pay more into their own pensions and lobbied for lower retirement benefits for newly hired workers.
In December, SEIU members protested outside the Mission Inn where Buster was holding a fundraiser. Signs called Buster ?the Grinch?.
